A buffer is a solution that neutralizes pH despite the presence of small amounts of an acid or base that may attempt to change the pH of a substance. The use of buffers is particularly significant in processes and reactions that require a stable pH. Buffers consist of a weak conjugate acid-base pair--meaning a weak acid and its conjugate base or a weak base and its conjugate acid. The combined components create the presence of an equilibrium which in turn balances the pH of the substance. The composition of the buffer is dependent on the desired pH of the process being tested or studied.

Buffers used specifically in biological research were studied and popularized by Norman Good, et al., and are commonly referred to today as Good buffers. These organic buffering compounds and solutions provide maximum buffering capacity between pH 6 and 8. Their use was a breakthrough in 1966 when biological research was previously stunted by reactive and even toxic buffers. Some of the more commonly used Good Buffers, in order of increasing pKa, include MES, ADA, PIPES, ACES, cholamine chloride, BES, TES, HEPES, acetamidoglycine, tricine, glycinamide, and Bicine. These buffers were selected on the basis of particular criteria including pH, solubility, membrane impermeability, low ionic/salt effects, low cationic complex activity, stability, optical absorbance, and ease of preparation. They also help maintain a nearly ideal pH state and are perfect for applications that require rigid regulation of buffer composition.
