Reagents in Protein Purification
The key to successful protein purification is choosing the correct process and the correct media and reagents. Although purification can sometimes be a simple one-step precipitation, multiple step processes are more frequently required to attain the desired purity. Your desired purity level will depend on the function to be served by the purified protein. Proteins for therapeutic use and in vivo studies generally require the highest purity level (> 99%), some characterization methods may require purity on the order of 95%-99%, while certain antibody production and sequencing methods may require just < 95% purity.

Additionally, your purification process will depend largely on the properties of your desired protein and targeted contaminants. Reagents and techniques should be chosen according to your objective in the corresponding step of your purification process, whether the aim is to isolate or purify the target protein.

For all steps in extracting, isolating and purifying proteins, protease inhibitors such as pepstatin, bestatin (structure below), leupeptin, E-64, AEBSF for aspartic proteases or ETDA for metal-dependent proteases can be used in the buffer solution to inhibit protease bestatin, protease inhibitordevelopment and prevent damage to the target protein.
