Description

Dithiobutylamine (DTBA) is a dithiol that can be synthesized from L-aspartic acid (A790021). Dithiobutylamine is a potent and versatile disulfide-reducing agent that is capable of reducing the enzyme papain, which has a relatively anionic active site, 14 times as fast as DTT could.

DTBA has thiol pKa values that are approximately 1 unit lower than DTT and forms a disulfide with a similar E value. Dithiobutylamine reduces disulfide bonds faster than DTT, especially in neutral aqueous systems. The amino group of DTBA enables its isolation by cation exchange and facilitates its conjucation.

Dithiobutylamine has very desirable physicochemical attributes. The HCl salt of DTBA is nearly odorless with a high solubility in water.

Dithiobutylamine is a new and improved reagent for the reduction of disulfide bonds in biological systems. The presence of the primary amine group in DTBA, as opposed to the two hydroxyl groups of DTT, leads to lower pKa values for the two thiols of the new reagent, attributing to its enhanced biological reducing activity. DTBA is simply the best reagent for reducing disulfide bonds in aqueous solutions. The HCl salt of DTBA is nearly odorless with a high solubility in water.
