Description

Methotrexate is an antineoplastic antimetabolite that is known to inhibit tetrahydrofolate dehydrogenase. Through this mechanism, methotrexate blocks DNA synthesis by preventing the formation of tetrahydrofolic acid. The compound is also known to possess considerable immunosuppressive properties, though the mechanism by which methotrexate suppresses immune responses is not fully understood.

Applications

Methotrexate is used in clinical medicine as a treatment for adult rheumatoid arthritis and for cases of severe psoriasis in which more standard therapies prove ineffective. It is also a widely used anticancer drug and is often administered in conjunction with other chemotherapeutic agents in cases of breast, lung and epidermoid cancers, as well as in the treatment of non-Hodgkin's lymphoma. Research into methotrexate has principally focused on its medical applications and long-term safety for patients. Other studies have attempted to investigate its immunosuppressive mechanisms.
