A potent and specific inhibitor of the protein kinase c-Raf (IC = 70 nM). Inhibits c-Raf with ten-fold increased potency compared to B-Raf, but does not inhibit many other protein kinases (even at 50 M) with the exception of SAPK2a/p38a (IC50 = 2 M) and SAPK2b/p38b2 (IC = 2 M). The inhibition of c-Raf by ZM 336372 is competitive with respect to ATP.
