Description

Thiazovivin is a Rho-associated kinase inhibitor which acts on the ROCK signaling pathway known to mediate several key cellular functions, including gene expression. Thiazovivin, sometimes abbreviated to TZV, is cell-permeable and has been put to wide use in studies of cell differentiation and transition because of its inhibitory effect on the ROCK pathway.

Applications

Research has shown that this inhibitor supports the survival and renewal of stem cells in both human and animal subjects. At least one human study has demonstrated thiazovivin's ability to inhibit the endothelial-to-mesenchymal and epithelial-to-mesenchymal transitions which are associated with corneal disease.

TZV has also been used in experiments relating to chemical reprogramming of cells. In one prominent example, thiazovivin was applied to porcine embryonic fibroblasts in conjunction with the TGF-beta growth factor SB431542, causing the PEFs to be reprogrammed as apidocytes.

Rho-associated kinase (ROCK) is a serine-threonine kinase known to play an important role in Rho-mediated cytoskeleton remodeling and contractile force generation. Down-regulation of ROCK signaling has been shown to enable the cell-adhesion interactions necessary for maintaining human embryonic stem cell survival and self-renewal.1 Thiazovivin is a selective, cell-permeable small molecule that inhibits ROCK activity. It promotes the survival of induced pluripotent stem cells and improves their reprogramming efficiency from human fibroblasts more than 200 fold when used in combination with the ALK5 inhibitor SB 431542 (Item No. 13031) and the MEK inhibitor PD0325091 (Item No. 13034).
