Ascomycin is an analog of FK506 with potent immunosuppressive antibiotic properties. It is an intensive calcium-dependent serine/threonine protein phosphatase inhibitor.
Ascomycin suppresses T-cell proliferation through binding to calcineurin (CaN) and interferes with dephosphorylation of various substrates. The mechanism downregulates production of interleukin 2 (IL2), interleukin 4 (IL4), and the release of cytokines.
It has known anticonvulsant activity in rat models. Because of CaN-mediated regulation of gamma aminobutyric acid (GABA) and glutamate receptor channels, ascomycin is related to neuronal cytoskeleton and dendritic spine morphology and inflammatory responses in gila cells. It blocks signaling pathways in astrocytes and changes the pattern of cytokine and neurotrophin gene expression.
Isolated from Streptomyces hygroscopicus var ascomyceticus.
