Blasticidin S HCl is an antibiotic that demonstrates a strong inhibitory effect on protein synthesis in eukaryotic and prokaryotic cells.

Blasticidin causes tRNA to bond more strongly to the P site of the large ribosomal subunit while simultaneously deforming the tRNA, thus inhibiting proper translation. The ultimate result of this process is a significant suppression of the hydrolysis of peptidyl-tRNA along with inhibition of peptide bond formation.

Isolated from Streptomyces griseochromogenes.
 

Aside from its antibiotic effects, blasticidin can be used to select for blasticidin resistance genes, which have undergone considerable study in recent years. One resistance gene, blasticidin S deaminase, has been used as a selection marker for genetic transformation and genome editing.

The antibiotic has also been used as a selection factor in studies of forced genetic expression in anticancer therapy, specifically of the CSR1 gene.
