TRIS-Tricine-SDS running buffer is used in the Schagger and Von Jagow method. This buffer is used to separate low molecular weight proteins and peptides. Peptide-SDS complexes move more slowly through Tris-tricine gels than they do through Tris-glycine buffers, allowing for the complete separation of SDS micelles that otherwise normally interfere with peptide separations. The tricine allows for efficient stacking and de-stacking of low molecular weight peptides, resulting in higher resolution of smaller peptides.
