Description

AEBSF HCl (also called Pefabloc) is a broad spectrum, water soluble, irreversible serine protease inhibitor. Inhibition constants are similar to those of PMSF and DFP. AEBSF has been shown to inhibit trypsin, chymotrypsin, plasmin kallikrein and thrombin. As an alternative to PMSF and DFP, AEBSF offers lower toxicity, improved solubility in water and improved stability in aqueous solutions. It has been shown to inhibit the constitutive production of beta amyloid (A beta) in five different human cell lines, both neural and non-neural.

AEBSF also inhibits activation of TGFβ (transforming growth factor beta), moderately inhibits dipeptidyl peptidase IV (DPP IV) and activates JNK and p38 MAP kinases. AEBSF is a b-secretase inhibitor that has been shown to inhibit beta amyloid peptide production and enhance APP (amyloid precursor protein) secretion in several cell lines at millimolar concentrations.

USE
The benzenesulfonyl fluoride derivative AEBSF is more stable in physiological medium than PMSF, is commercially available, and is widely used as a broad-spectrum serine protease inhibitor. AEBSF inhibits proteases like chymotrypsin, kallikrein, plasmin, thrombin, and trypsin. AEBSF also inhibits activation of TGFβ, DPP IV and activates JNK and p38 MAP kinases. AEBSF is a b-secretase inhibitor that has been shown to inhibit beta amyloid peptide production and enhance APP (amyloid precursor protein) secretion in several cell lines. AEBSF is extensively used in studies aiming to describe cholesterol regulatory genes due to its potent ability to inhibit Site-1-protease (S1P). This serine protease, located in the Golgi apparatus, is responsible for activating the sterol regulatory element-binding proteins (SREBP). By selectively inhibiting S1P, AEBSF can be used to characterize the downstream result of SREBP inhibition and its influence on cholesterol regulation.
