Vinblastine sulfate is a plant alkaloid, arresting the cell cycle in the G2/M phase by blocking mitotic spindle formation. It suppresses normal microtubule assembly and causes abnormal tubulin polymerization. It also inhibits binding of tubulin to microtubule proteins.
The plant alkaloid is related to the apoptosis pathway. It renders Raf-1 activation, phosphorylation of Bcl-2 family proteins, and induction of p53 expression. Apoptosis in human acute promyelocytic leukemia cells (APL) is induced by vinblastine sulfate because it inhibits nuclear factor kappa B (NFκB) expression and suppresses NFκB-DNA binding activity. Also, Induction of c-Jun N-terminal kinase (JNK) activation causes apoptosis through caspase-dependent pathway.
