Pladienolide B is the major analogue of a family of macrocyclic lactones isolated from Streptomyces platensis. Pladienolide B is a highly potent inhibitor of both hypoxia signals and cancer cell proliferation. Pladienolide B is a complex natural product that potently inhibits pre-mRNA splicing.
Pladienolide B binds to SF3b complex blocking the spliceosome.Pladienolide B analogs can be used to study the relationship between splicing and cancer cell function. RNA splicing as in attractive target for development of effective anticancer therapy including Lymphoma and Chronic Lymphocytic Leukemia.
